Effect of Deep Sea Water Containing Chitosan Oligosaccharides on Glucose Uptake in 3T3-L1 Adipocytes

Abstract
To investigate the effect of deep sea water (DSW) containing chitosan oligosaccharides (COS) on glucose uptake, we performed glucose uptake assay, RT-PCR and western blot in mature 3T3-L1 adipocytes with the treatment of DSW or DSW containing COS. Both DSW and DSW containing COS treatment increased glucose uptake in mature 3T3-L1 adipocytes. RT-PCR results revealed that the expressions of genes related to glucose uptake such as GLUT 4 for glucose transporter, IRAP for an abundant cargo protein of GLUT 4 storage vesicles (GSVs), Syntaxin 6, and Syntaxin16 for vesicle trafficking were increased. Moreover, the phosphorylation of IRS 1, which is an important signal molecule for insulin-dependent glucose uptake, was increased by DSW and DSW containing COS treatment in mature 3T3-L1 adipocytes. In conclusion, these results suggest that DSW containing COS is more effective than DSW alone in stimulatory effect on glucose uptake.
